Denis Zakharov began his basketball career in his hometown of Belgorod under the guidance of renowned coach Viktor Shved, the father of Alexey Shved. He started his professional career in the Russian Superleague with the club Sparta&K, where Zakharov emerged as one of the team's leaders. In May 2013, he joined Nizhny Novgorod and played there for a season before signing a contract with Dynamo Moscow, where he had successful performances for two seasons.
Following Dynamo's bankruptcy, Zakharov, who had already become a bronze medalist in the Superleague and the league's best shooting guard, moved to Enisey. He helped the team reach the FIBA Europe Cup Final Four and later signed a contract with Lokomotiv-Kuban in Krasnodar. After spending just a year with Lokomotiv-Kuban, he returned to Enisey, where he once again became one of the team's leaders.
In 2020, the guard moved to St. Petersburg and has been playing for Zenit ever since. In the 2021/22 season, he won the VTB United League together with Zenit.
